ABSTRACT

Objetivo: Analisar o efeito do consumo de diferentes fórmulas de café enriquecidas com cacau e canela sobre biomarcadores cardiovasculares, glicêmicos e antropométricos em mulheres hipertensas, durante 12 semanas. Métodos: Ensaio clínico randomizado duplo-cego, do tipo fatorial, a ser realizado em Unidades Básicas de Saúde, em uma cidade do interior do Ceará, com uma amostra de 90 pessoas. Os participantes elegíveis passarão por duas avaliações (antes e depois) para mensuração de biomarcadores cardiovasculares clínicos e laboratoriais, glicêmicos e antropométricos. A randomização será por estratos, de acordo com o estágio da hipertensão, e a alocação será por blocos. Os participantes serão orientados a tomar as fórmulas de café duas vezes ao dia (café da manhã e almoço), em concentrações de 10g (café), 5g (cacau) e 3g (canela), preparados com 50 mL de água quente. Para análise dos dados, será considerada a ANOVA de medidas repetidas (> 2 grupos), e em caso de associações estatisticamente significantes (P< 0,05), será realizada regressão linear. Com isso, pretende-se conhecer qual é a fórmula de café enriquecido mais eficaz para o controle e/ou redução de biomarcadores cardiológicos, glicêmicos e antropométrico. Objective: To analyze the effect of consuming different coffee formulas enriched with cocoa and cinnamon on cardiovascular, glycemic and anthropometric biomarkers in hypertensive women, during 12 weeks.Methods:Double-blind randomized clinical trial, of the factorial type, to be carried out in Basic Health Units, in a city in the interior of Ceará, with a sample of 90 people. Eligible participants will undergo two assessments (before and after) to measure clinical and laboratory cardiovascular, glycemic and anthropometric biomarkers. Randomization will be by strata, according to the stage of hypertension, and allocation will be by blocks. Participants will be instructed to drink coffee formulas twice a day (breakfast and lunch), in concentrations of 10g (coffee), 5g (cocoa) and 3g (cinnamon), prepared with 50 mL of hot water. For data analysis, repeated measures ANOVA (> 2 groups) will be considered, and in case of statistically significant associations (P< 0.05), linear regression will be performed. With this, it is intended to know which is the most effective fortified coffee formula for the control and/or reduction of cardiological, glycemic and anthropometric biomarkers. ABSTRACT

Introducción. El síndrome metabólico (SM) aumenta el ingreso hospitalario y el riesgo de desarrollar COVID-19, los fármacos utilizados para su tratamiento ocasionan efectos secundarios por lo que se ha optado por la búsqueda de alternativas terapéuticas a base de compuestos bioactivos contenidos en plantas medicinales. La canela se utiliza como agente terapéutico debido a sus propiedades comprobadas con diversos mecanismos de acción reportados en el tratamiento de varias patologías. Objetivo. Documentar los estudios in vitro, in vivo, estudios clínicos y los mecanismos de acción reportados del efecto de la administración de extractos y polvo de canela en las comorbilidades relacionadas con el SM. Materiales y métodos. Revisión sistemàtica de artículos en bases de datos electrónicas, incluyendo estudios de canela en polvo, extractos acuosos, de acetato de etilo y metanol de la corteza de canela, período de 5 años, excluyendo todo artículo relacionado a su efecto antimicrobiano, antifúngico y aceite de canela. Resultados. Las evidencias de los principales compuestos bioactivos contenidos en la canela validan su potencial en el tratamiento de enfermedades relacionadas al SM, con limitados estudios que indagan en los mecanismos de acción correspondientes a sus actividades biológicas. Conclusiones. Las evidencias de las investigaciones validan su potencial en el tratamiento de estas patologías, debido a sus principales compuestos bioactivos: cinamaldehído, transcinamaldehído, ácido cinámico, eugenol y, antioxidantes del tipo proantocianidinas A y flavonoides, los cuales participan en diversos mecanismos de acción que activan e inhiben enzimas, con efecto hipoglucemiante (quinasa y fosfatasa), antiobesogénico (UPC1), antiinflamatorio (NOS y COX), hipolipemiante (HMG-CoA) y antihipertensivo (ECA)(AU)


Introduction. Metabolic syndrome (MS) increases hospital admission and the risk of developing COVID-19. Due to the side effects caused by the drugs used for its treatment, the search for therapeutic alternatives based on bioactive compounds contained in medicinal plants has been chosen. Cinnamon is used as a therapeutic agent due to its proven properties with various mechanisms of action reported in the treatment of various pathologies. Objective. To document the in vitro and in vivo studies, clinical studies and the mechanisms of action reported on the effect of the administration of cinnamon extracts and powder on comorbidities related to MS. Materials and methods. Systematic review of articles in electronic databases, including studies of cinnamon powder, aqueous extracts, ethyl acetate and methanol from cinnamon bark, over a period of 5 years, excluding all those articles related to its antimicrobial, antifungal and antimicrobial effect. cinnamon oil. Results. The evidence of the main bioactive compounds contained in cinnamon validates its potential in the treatment of diseases related to MS, with limited studies that investigate the mechanisms of action corresponding to its biological activities. Conclusions. Research evidence validates its potential in the treatment of these pathologies, due to its main bioactive compounds: cinnamaldehyde, transcinnamaldehyde, cinnamic acid, eugenol, and antioxidants of the proanthocyanidin A type and flavonoids, which participate in various mechanisms of action that activate and they inhibit enzymes, with hypoglycemic (kinase and phosphatase), antiobesogenic (UPC1), anti-inflammatory (NOS and COX), lipid-lowering (HMG-CoA) and antihypertensive (ACE) effects(AU)

ABSTRACT

A resistência bacteriana tem aumentado progressivamente no mundo, assim, há necessidade de novas opções de tratamentos. A fitoterapia tem ganhado notoriedade para combater infecções, principalmente as causadas por bactérias resistentes aos antibacterianos disponíveis. Diante do exposto, o presente estudo teve como objetivo preparar e analisar a composição fitoquímica e a ação antibacteriana dos extratos hidroetanólicos de canela (EHC) e romã (EHR) isolados e associados frente culturas planctônicas e biofilmes de cepas padrão e clínicas de Acinetobacter baumannii e Pseudomonas aeruginosa, além disso, analisar a ação citotóxica dos extratos em queratinócitos humanos (HaCat). Para isso, os EHC e EHR foram preparados e quantificado o teor de sólidos solúveis. Posteriormente, foi quantificado o teor de flavonoides e fenóis totais, análise antioxidante por meio da redução do radical 2,2'-difenil-1-picrilhidrazila (DPPH), e a fitoquímica por cromatografia líquida (HPLC). Em relação a ação antibacteriana dos extratos, foi aplicado o teste de microdiluição em caldo (CLSI ­ M7-A9) e a ação sinérgica realizada por meio do ensaio de checkerboard. As concentrações mais efetivas foram analisadas sobre biofilmes em formação (prevenção) e biofilmes formados (tratamento de 24 h), e quantificada a viabilidade por meio do teste colorimétrico MTT. Para avaliar a citotoxidade, os tratamentos foram aplicados sobre cultura celular de HaCat por 24 h e analisados por meio do teste colorimétrico MTT. A análise estatística foi realizada com 5% de significância (p<0.05), analisados pelo método ANOVA complementado pelo Teste de Tukey. Os resultados demonstraram que os EHC e EHR possuem ação antioxidante e presença de fitocompostos. Os extratos apresentaram ação antibacteriana para todas as cepas avaliadas, quando os mesmos foram associados, obteve-se concentrações sinérgicas para as cepas clínicas de A. baumannii. Em relação a ação antibiofilme, o EHC inibiu a formação em 95% e EHR em 96% do biofilme de #Ab 1, enquanto a cepa #Pa 2 teve 92% e 93% de inibição quando em contato com EHC e EHR, respectivamente. Após tratamento de 24 h em biofilmes formados, as reduções da viabilidade foram de 72% para as cepas #Ab 2 e #Ab 3 quando em contato com o EHC, já EHR inibiu em 83% a viabilidade da cepa #Ab ATCC. Para P. aeruginosa (#Pa 2), as reduções da viabilidade foram de 84% e 88,5% quando tratados com EHC e EHR, respectivamente. A avaliação da citotoxicidade em HaCat demonstrou que após tratamentos com diferentes concentrações dos extratos a viabilidade celular se manteve acima de 70% em todos os grupos. Diante disso, conclui-se que os EHC e EHR apresentam importante ação antioxidante e antibacteriana, tanto em culturas planctônicas quanto em biofilmes, e não apresentaram efeitos citotóxicos na faixa de concentração testada. Bacterial resistance has progressively increased in the world, thus, there is a need for new treatment options. Phytotherapy has gained notoriety for fighting infections, mainly those caused by bacteria resistant to available antibacterials. In view of the above, the present study aimed to prepare and analyze the phytochemical composition and antimicrobial action of hydroethanolic extracts of cinnamon (EHC) and pomegranate (EHR) isolated and associated against planktonic cultures and biofilms of standard and clinical strains of Acinetobacter baumannii and Pseudomonas aeruginosa, in addition, analyze the cytotoxic action of the extracts on human keratinocytes (HaCat). For this, the EHC and EHR were prepared and the soluble solids content was quantified. Subsequently, the content of flavonoids and total phenols, antioxidant analysis through the reduction of the radical 2,2'-diphenyl1-picrylhydrazyl (DPPH), and phytochemistry by liquid chromatography (HPLC) were quantified. Regarding the antimicrobial action of the extracts, the broth microdilution test (CLSI ­ M7-A9) was applied and the synergistic action was performed through the checkerboard test. The most effective concentrations were analyzed on forming biofilms (prevention) and formed biofilms (24 h treatment), and viability was quantified using the MTT colorimetric test. To evaluate the cytotoxicity, the treatments were applied on HaCat cell culture for 24 h and analyzed using the MTT colorimetric test. Statistical analysis was performed with 5% significance (p<0.05), analyzed by the ANOVA method complemented by the Tukey test. The results showed that the EHC and EHR have antioxidant action and presence of phytocompounds. The extracts showed antibacterial action for all evaluated strains, when they were associated, synergistic concentrations were obtained for the clinical strains of A. baumannii. Regarding the antibiofilm action, EHC inhibited formation by 95% and EHR by 96% of the #Ab 1 biofilm, while the #Pa 2 strain had 92% and 93% inhibition when in contact with EHC and EHR, respectively. After 24 h treatment in formed biofilms, viability reductions were 72% for strains #Ab 2 and #Ab 3 when in contact with EHC, whereas EHR inhibited the viability of strain #Ab ATCC by 83%. For P. aeruginosa (#Pa 2), viability reductions were 84% and 88.5% when treated with EHC and EHR, respectively. The evaluation of cytotoxicity in HaCat showed that after treatments with different concentrations of extracts, cell viability remained above 70% in all groups. Therefore, it is concluded that EHC and EHR have important antioxidant and antibacterial action, both in planktonic cultures and in biofilms, and did not show cytotoxic effects in the tested concentration range. Introduction: some plants such as turmeric, cinnamon, and okra are known to have therapeutic functions such as antioxidant and anti-inflammatory activity. Furthermore, an immunomodulatory role has been observed in the production of antibodies, in particular immunoglobulin A (IgA), which mediates a variety of protective functions for the organism. Objective: the aim of the present study was to investigate the effect of dietary plants on the production of IgA in healthy Wistar rats. Methods: thus, 48 male Wistar rats of 90 days of age were allocated to four groups. The animals were treated for 14 days with dried turmeric, cinnamon, or okra (50, 50, 12.5 mg/day, respectively) in phosphate buffered saline, or with only phosphate buffered saline by gavage. The animals received water and feed ad libitum. Body mass and relative weight ofperitoneal fat, adrenal gland, kidney, spleen, liver and thymus, biochemical parameters, and IgA levels were analyzed. Results: no significant changes were observed in the body mass, relative weight of organs and tissues, and biochemical parameters. An increase in serum IgA levels was observed in animals treated with turmeric or cinnamon. Conclusion: we conclude that the treatment with turmeric and cinnamon increased IgA production. Therefore, our study supports the idea that dietary supplementation with these plants may improve humoral immunity.


Introdução: algumas plantas como a cúrcuma, a canela e o quiabo são conhecidas por apresentar funções terapêuticas, como atividade antioxidante e anti-inflamatória. Além disso, tem sido observado um papel imunomodulador sobre a produção de anticorpos, em especial a imunoglobulina A (IgA), a qual medeia uma variedade de funções protetoras para o organismo. Objetivo: o objetivo do presente estudo foi investigar o efeito de plantas dietéticas na produção de IgA em ratos Wistar saudáveis. Métodos: destarte, 48 ratos machos Wistar com 90 dias de idade foram alocados em quatro grupos. Os animais foram tratados por 14 dias com cúrcuma seca, canela ou quiabo (50, 50, 12,5 mg/dia, respectivamente) em solução salina tamponada com fosfato ou apenas solução salina tamponada com fosfato, por gavagem. Os animais receberam água e ração ad libitum. Foram analisados a massa corporal e o peso relativo da gordura peritoneal, glândula adrenal, rim, baço, fígado e timo, parâmetros bioquímicos e níveis de IgA. Resultados: não foram observadas alterações significativas na massa corporal, no peso relativo dos órgãos e tecidos e nos parâmetros bioquímicos. Foi observado aumento dos níveis séricos de IgA nos animais tratados com cúrcuma ou canela. Conclusão: podemos concluir que o tratamento com cúrcuma e canela aumentou a produção de IgA. Portanto, nosso estudo suporta a ideia de que a suplementação alimentar com essas plantas pode melhorar a imunidade humoral.

This study was aimed to explore the comparative efficacy of cinnamon bark extract, cinnamaldehyde and kaempferol against acetaminophen (APAP)-induced oxidative stress. Cinnamon bark extract, cinnamaldehyde and kaempferol were utilized or in-vivo analysis. From the results of in-vitro screening tests, cinnamon ethanolic extract was selected for in-vivo study in mouse model. For this, Balb/c albino mice were treated with cinnamon ethanolic extract (200 mg/kg), cinnamaldehyde (10 mg/kg) and kaempferol (10 mg/kg) orally for 14 days followed by single intraperitoneal administration of APAP during 8 hours. Blood and organ samples were collected for biochemical and histopathological analysis. The results showed that cinnamon bark ethanolic extract, cinnamaldehyde and kaempferol ameliorated APAP-induced oxidative stress and organ toxicity in mice. In conclusion, cinnamaldehyde and kaempferol possess comparable antioxidant potential even at 20-times less dose as compared to cinnamon bark ethanolic extract suggesting therapeutic potential in oxidative stress-related disorders.


Este estudio tuvo como objetivo explorar la eficacia comparativa del extracto de corteza de canela, cinamaldehído y kaempferol contra el estrés oxidativo inducido por acetaminofén (APAP). Se utilizaron extracto de corteza de canela, cinamaldehído y kaempferol para el análisis in vivo. De los resultados de las pruebas de detección in vitro, se seleccionó el extracto etanólico de canela para estudio in vivo en modelo de ratón. Para ello, los ratones albinos Balb/c fueron tratados con extracto etanólico de canela (200 mg/kg), cinamaldehído (10 mg/kg) y kaempferol (10 mg/kg) por vía oral durante 14 días, seguido de la administración intraperitoneal única de APAP durante 8 horas. Se recogieron muestras de sangre y órganos para análisis bioquímicos e histopatológicos. Los resultados mostraron que el extracto etanólico de la corteza de canela, el cinamaldehído y el kaempferol mejoraron el estrés oxidativo inducido por APAP y la toxicidad orgánica en ratones. En conclusión, el cinamaldehído y el kaempferol poseen un potencial antioxidante comparable, incluso a una dosis 20 veces menor en comparación con el extracto etanólico de la corteza de canela, lo que sugiere un potencial terapéutico en los trastornos relacionados con el estrés oxidativo.

ABSTRACT

The liver as an organ is important for the metabolism of drugs and toxins. However, it is not immune from environmental insults. Exposure of liver cells to carbon tetrachloride (CCl4) results in the generation of tricholoromethyl radicals, which induce liver toxicity. This study aims at investigating the ameliorative effect of the cinnamon aqueous extract (CAE) against CCl4-induced hepatotoxicity in male albino rats. Hepatotoxicity was induced in rats through the intraperitoneal administration of 0.5 mL kg-1body weight of CCl4. The analyses of the results obtained showed significant reduction in the levels of serum biochemical markers for 400 and 600 mg kg-1bw of CAE protected rats as compared with CCl4group. In addition, CAE administration reversed liver tissue damaged via increased antioxidants markers. Histopathological examination of CAE treatment on rats showed improved changes to the liver damage caused by CCl4 with no evidence of steatosis and inflammation. This result hence suggests that CAE has marked hepatoprotective and healing activities against CCl4-induced liver damage and could serve as a suitable candidate in drug discovery for the treatment of liver toxicity.

Abstract This study aimed to chemically characterize the essential oils (EOs) of Cinnamomum zeylanicum (cinnamon) and Eremanthus erythropappus (candeia) and evaluate their acaricidal activity, together with that of their major compounds and cinnamyl acetate derivative, against Rhipicephalus microplus. Essential oil compounds were identified through gas chromatography. The larval packet test (LPT) at concentrations ranging from 0.31 to 10.0 mg/mL and the adult immersion test (AIT) at concentrations between 2.5 and 60.0 mg/mL were performed. (E)-cinnamaldehyde and α-bisabolol were the major compounds in cinnamon (86.93%) and candeia (78.41%) EOs, respectively. In the LPT, the EOs of cinnamon and candeia and the compounds (E)-cinnamaldehyde, α-bisabolol and cinnamyl acetate resulted in 100% mortality at concentrations of 2.5, 2.5, 5.0, 10.0 and 10.0 mg/mL respectively. In the AIT, percentage control values > 95% were observed for cinnamon and candeia EOs, (E)-cinnamaldehyde and α-bisabolol at the concentrations of 5.0, 60.0, 20.0, and 20.0 mg/mL, respectively, whereas cinnamyl acetate showed low activity. We conclude that EOs and their compounds showed high acaricidal activity, whereas the acetylated derivative of (E)-cinnamaldehyde presented less acaricidal activity on R. microplus engorged females.


Resumo Este estudo teve como objetivo caracterizar quimicamente os óleos essenciais (OE) de Cinnamomum zeylanicum (canela) e Eremanthus erythropappus (candeia) e avaliar sua atividade acaricida, juntamente com a de seus principais compostos e do derivado de acetato de cinamila, sobre Rhipicephalus microplus. Os compostos do óleo essencial foram identificados por cromatografia gasosa. Foram realizados o Teste de Pacote de Larvas (TPL), em concentrações variando de 0,31 a 10,0 mg/mL, e o Teste de Imersão de Adultos (TIA), em concentrações entre 2,5 e 60,0 mg/mL. (E)-cinnamaldeído e α-bisabolol foram os principais compostos nos OE da canela (86,93%) e da candeia (78,41%), respectivamente. No TPL, os OEs de canela e candeia, e os compostos (E)-cinnamaldeído, α-bisabolol e acetato de cinamila resultaram em 100% de mortalidade nas concentrações de 2,5, 2,5, 5,0, 10,0 e 10,0 mg/mL, respectivamente. No TIA, valores percentuais de controle >95% foram observados para OE de canela e candeia, (E)-cinnamaldeído e α-bisabolol nas concentrações de 5,0, 60,0, 20,0 e 20,0 mg/mL, respectivamente, enquanto o acetato de cinamila apresentou baixa atividade. Conclui-se que os OEs e seus compostos apresentaram alta atividade acaricida, enquanto o derivado acetilado do (E)-cinnamaldeído apresentou menor atividade acaricida em fêmeas ingurgitadas de R. microplus.

ABSTRACT

O objetivo deste estudo foi avaliar a atividade antifúngica dos óleos essenciais de Cinnamomum cassia (L.) J.Presl, Myristica fragrans Houtt, Schinus terebinthifolius Raddi, Zingiber officinale Roscoe bem como o composto químico majoritário do óleo essencial com a menor concentração fungicida mínima sobre 16 isolados biológicos de Candida albicans. Determinar as concentrações fungicidas mínimas, o efeito dos óleos e do composto químico majoritário, em doses subinibitórias sobre a formação de tubo germinativo, clamidoconídeo e a produção de franjas, nos isolados de C. albicans, antes e após contato com os diferentes óleos essenciais e composto químico majoritário. Os quatro óleos essenciais apresentaram atividade inibitória sobre C. albicans. C. cassia (L.) J.Presl CFM 50 3,23 µg/mL e CFM 90 51,68 µg/mL ,M. fragrans Houtt CFM 50 21,437 µg/mL e CFM 90 171.500 µg/mL, S. terebinthifolius Raddi CFM 50 42.600 µg/mL e CFM 90 85.200µg/mL, Z. officinale Roscoe CFM 50 171.200µg/mL e CFM 90 resistente até a concentração de 171.200 µg/mL, sendo o óleo essencial de C. cassia (L.) J.Presl o que apresentou a menor concentração fungicida sobre os isolados de C. albicans. Nas doses subinibitórias os óleos essenciais de C. cassia (L.) J.Presl (canela), M. fragans Houtt (noz moscada), S. terebinthifolius Raddi (pimenta rosa), Z. officinale Roscoe (gengibre) não inibiram em todos os isolados a formação de tubo germinativo e clamidoconídeo, mas alteraram a produção de franjas. O composto químico majoritário aldeído cinâmico do óleo essencial de C. cassia (L.) J.Presl com CFM 50 26,64µg/mL e CFM 90 213,18 µg/mL, apresentou atividade fungicida sobre os isolados de C. albicans, mas pouca atividade inibitória na formação de tubo germinativo, clamidoconídeo e produção de franja. O óleo essencial de C. cassia (L.) J.Presl (canela), apresentou as melhores concentrações fungicidas mínimas e inibição da formação de tubo germinativo em relação ao composto químico majoritário aldeído cinâmico.

ABSTRACT

Abstract The aim of this study was to evaluate the antimicrobial effect of the essential oils of cinnamon, cardamom, clove, oregano, and thyme and their synergism on vegetative cells and endospores of Clostridium perfringens type A inoculated in meat sausage (mortadella), as well as the influence of blends on the color, and lipid oxidation through the determination of thiobarbituric acid reactive substances (TBARS index). The anticlostridial action of the oil blends was established. The two added oil blends (Treat. 1: oregano, clove, and thyme; Treat. 2: oregano, clove, and cinnamon) in combination with reduced nitrite content (75 ppm) promoted a lower growth of C. perfringens in mortadella stored at 15 °C for 21 days in comparison to treatments containing only 75 ppm of nitrite. The essential oil blends showed antioxidant action and did not alter food color, thus possessing potential application as a preservative for the meat products industry.

ABSTRACT

Hot melt pressure-sensitive adhesive(HMPSA) has broad application potential in the field of traditional Chinese medicine(TCM) plasters due to its high drug loading, weak skin irritation, satisfactory adhesion, etc. compared with rubber plasters.However, the structure of HMPSA is prone to suffer from the damage caused by volatile oils in TCM plasters. In view of this, a kind of HMPSA with a stable structure was prepared by physical blending of DINCH, polypropylene wax and liquid rubber(LIR) in the present study, which is denoted as DPL. The dosage of cinnamon volatile oil(CVO), the model drug, was selected with viscosity, softening point and cohesion as evaluation indexes. The interaction between DPL and HMPSA was investigated by Fourier transform infrared spectroscopy(FT-IR) and differential scanning calorimetry(DSC). The compatibility of HMPSA with CVO and its transdermal ability were studied by in vitro transdermal test, adhesion, scanning electron microscopy( SEM) and rheological evaluation. The results showed that 5% CVO began to damage the structure of HMPSA. The initial adhesion and holding adhesion of DPL-modified HMPSA(DPL-HMPSA) were not significantly changed compared with those of HMPSA, whereas the 180° peel strength was decreased. FI-IR unraveled that DPL formed the n-π conjugated system with styrene-isoprene-styrene block copolymer(SIS), and there was no significant difference in the glass transition temperature according to DSC results, which indicated the good compatibility of DPL with HMPSA. With 5% CVO loaded, the drug content of DPL-HMPSA was 1. 14 times higher than that of HMPSA, and the decrease rate of drug content in DPL-HMPSA was 16% lower than that in HMPSA after 3 months. SEM demonstrated that CVO did not cause obvious structural damage to DPL-HMPSA. Rheological evaluation revealed that the storage modulus and loss factor of DPL-HMPSA were higher than those of HMPSA, and the cohesion was also stronger. The percutaneous penetration rate of cinnamaldehyde in DPL-HMPSA was 2. 25 times that of HMPSA. In conclusion, DPL-HMPSA had more stable structure, better compatibility with CVO, and higher in vitro transdermal efficiency of cinnamaldehyde than before the modification. This study can provide reference for the mitigation of the matrix structure damage caused by volatile oil components in TCM plasters and the enhancement of the content and in vitro transdermal rate of drug.

ABSTRACT

Este estudo teve como objetivo analisar a eficácia da canela (Cinnamomum verum) na redução dos níveis glicêmicos de pessoas com Diabetes Mellitus do tipo 2, em comparação ao placebo. Trata-se de um ensaio clínico aleatorizado, triplo cego, controlado por placebo, desenvolvido em cinco Unidades Básicas de Saúde, na cidade de Parnaíba ­ PI. A população do estudo foi constituída de adultos e idosos, de ambos os sexos, com diagnóstico para Diabetes Mellitus do tipo 2. Após os devidos cálculos, o tamanho amostral foi de 154 pessoas. Os critérios de inclusão foram: pessoas em uso de antidiabéticos orais, com HbA1c ≥ 6,0%; e os critérios de exclusão: pessoas com alergia à canela, em uso de insulina ou com diagnóstico de complicações severas que pudessem comprometer a análise. Os participantes foram distribuídos em dois grupos (experimental e controle) e aleatorizados por um programa de computador. A alocação foi feita de 1:1, em grupos paralelos, de acordo com os valores de HbA1c. Os pacientes do grupo experimental receberam frascos contendo cápsulas com canela e foram orientados a tomar 3 g por dia, durante 90 dias. Os do grupo controle receberam frascos contendo cápsulas idênticas às do grupo experimental, mas com celulose microcristalina (placebo), sendo orientados a tomar a mesma quantidade de cápsulas por dia, durante 90 dias. Variáveis socioeconômicas, clínicas, relacionadas ao estilo de vida, laboratoriais e antropométricas foram avaliadas. O test t foi utilizado para comparar as médias das variáveis numéricas e o teste quiquadrado foi utilizado para comparar as distribuições nas categorias dos dois grupos. A amostra final foi de 140 pessoas, sendo a maioria composta por participantes do sexo feminino (69%), de cor parda (57%) e média de idade de 60 anos. Após 90 dias, a média de glicemia venosa de jejum diminuiu 10 mg/dL no grupo experimental e aumentou 21 mg/dL no grupo controle (p= 0,001). Os valores de HbA1c caíram 0,2% no grupo canela e aumentaram 0,4% no grupo placebo (p= 0,001). Reduções estatisticamente significantes também foram observadas nos níveis de triglicerídeos e índice HOMA-IR, no grupo experimental. A canela, na concentração ofertada, mostrou-se eficaz para reduzir os níveis glicêmicos. No entanto, ainda não é possível fazer generalizações, visto que as análises realizadas podem ser influenciadas por inúmeras variáveis. Recomenda-se que outros estudos sejam realizados, bem como a avaliação do custo-efetividade da intervenção. (AU)

La presente investigación aborda el campo clínico del comportamiento de las cepas del hongo candida albicans, causantes de infecciones oportunistas en pacientes inmuno suprimidos, así como de las alternativas terapéuticas empleando medicina natural (Aceite de Canela) así como de fármacos (Nistatina), en el tratamiento anti fúngico. En el Capítulo I, se hace referencia al planteamiento del problema, señalando la formulación del mismo, los objetivos y la trascendencia de la investigación. El Capítulo II se hace referencia al marco teórico, precisando los conocimientos teóricos, epistemológicos, enfoques, teorías, modelos y fundamentos paradigmáticos correspondientes a la problemática evidenciada en los hallazgos de laboratorio respecto a las cepas y sus efectos en el ser humano. En el Capítulo III se señala el marco metodológico indicando el tipo de investigación, población, muestra; así mismo de las técnicas e instrumentos de recolección de datos, procesamiento y análisis de la información obtenida durante el desarrollo de la investigación. En el Capítulo IV se señalan los resultados o hallazgos de la investigación; finalmente en el Capítulo V se precisa la discusión y contrastación de los resultados. El estudio concluyó en que existen diferencias significativas entre el aceite de canela y la nistatina, empleadas en el tratamiento anti fungicida de la candida albicans, en concentraciones al 50% y 100%, pero no existen diferencias significativas diferencias significativas al 25% del aceite de canela.

BACKGROUND Essential oils (EO) extracted from Cinnamomum verum has been used as an antimicrobial agents for centuries. The effects of C. verum leaf oil against virulence of microorganisms is not well studied yet. OBJECTIVES This study evaluates the effect of C. verum leaf oil against three virulence factors of Candida albicans, C. tropicalis and C. dubliniensis and its in-vivo toxicity. METHODS Chemical composition of EO was determined using gas chromatography-mass spectrometry (GC-MS). Minimum inhibitory concentration (MIC) was determined using clinical and laboratory standards institute (CLSI) M27-A3 broth microdilution. Effect of EO on initial adhesion was quantified using XTT assay after allowing Candida cells to adhere to the polystyrene surface for 2 h. Biofilm formation of Candida in the presence of EO was quantified using XTT viability assay. Efficacy on reduction of germ tube formation was evaluated using standard protocol. Visualisation of biofilm formation and progression under the EO treatment were done using scanning electron microscope (SEM) and Time lapses microscope respectively. In-vivo toxicity of EO was determined using Galleria mellonella larvae. Chlorhexidine digluconate: positive control. RESULTS Eugenol was the main compound of EO. MIC was 1.0 mg/mL. 50% reduction in initial adhesion was achieved by C. albicans, C. tropicalis and C. dubliniensis with 1.0, > 2.0 and 0.34 mg/mL respectively. 0.5 and 1.0 mg/mL significantly inhibit the germ tube formation. MBIC50 for forming biofilms were ≤ 0.35 mg/mL. 1.0 mg/mL prevent biofilm progression of Candida. SEM images exhibited cell wall damages, cellular shrinkages and decreased hyphal formation. No lethal effect was noted with in-vivo experiment model at any concentration tested. CONCLUSION C. verum leaf oil acts against virulence factors of Candida and does not show any toxicity.

As an important edible herb and export variety of China, the quality control and internationalization of quality standards of C. cassis is extremely significant. In the recent years, with the development of the cinnamon industry, relevant academic research and the upgrade of the international standards, it is necessary to summarize the quality-related progress of C. cassis. In the present review, the germplasm resources, specific quality marker(Q-marker) and quality standards of C. cassis were summarized on the basis of published research during the last 10 years.

ABSTRACT Cinnamomum cassia (Cinnamon) is a well-known traditional medicine with therapeutic benefits for centuries. We evaluated the effects of cinnamon essential oil (CEO) and its main component cinnamaldehyde (CA) on human corpus cavernosum (HCC) and rat CC. The essential oil of cinnamon was analyzed for the confirmation of the oil profile. HCC specimens from patients undergoing penile prosthesis surgery (age 48-69 years) were utilized for functional studies. In addition, erectile responses in anesthetized control and diabetic rats were evaluated in vivo after intracavernosal injection of CEO and CA, and rat CC strips were placed in organ baths. After precontraction with phenylephrine (10µM), relaxant responses to CEO and CA were investigated. CA (96.9%) was found as the major component. The maximum relaxation responses to CEO and CA were 96.4±3.5% and 96.0±5.0% in HCC and 97.5±5.5% and 96.8±4.8% in rat CC, respectively. There was no difference between control and diabetic rats in relaxation responses to CEO and CA. The relaxant responses obtained with essential oil and CA were not attenuated in the presence of nitric oxide synthase (NOS) inhibitor, and soluble guanylate cyclase inhibitor (sGS) in CC. In vivo, erectile responses in diabetic rats were lower than in control rats, which was restored after intracavernosal injection of CEO and CA. CEO and CA improved erectile function and relaxation of isolated strips of rat CC and HCC by a NO/cGMP-independent mechanism. Further investigations are warranted to fully elucidate the restorative effects of CEO and CA on diabetic erectile dysfunction.

ABSTRACT

O objetivo desse trabalho foi analisar a qualidade microbiológica das especiariascanela, orégano e pimenta do reino comercializadas na feira livre de Cruz das Almas no Recôncavo da Bahia, por meio da quantificação de coliformes a 45°C, Staphylococcus spp., Staphylococcus coagulase positiva, Bacillus spp., Escherichia coli, Bacillus cereus e Salmonella spp. A pimenta do reino diferiu estatísticamente (p>0,05) apresentando maior contagem quando comparado com a canela e o orégano. A prevalência de S. coagulase positiva foi de 60%, 57% e 47% para pimenta do reino moída, orégano e canela, respectivamente. E. coli e Salmonella estiveram presentes na pimenta do reino em 94% e 20%, respectivamente. A contagem de coliformes a 45ºC na pimenta foi maior (3,4 log NMP. g-1) em relação a canela (0,5 log NMP.g-1) e orégano (ausência). A pimenta do reino moída apresentou alta carga microbiana com patógenos de importância alimentar como Salmonella spp., Staphylococcus coagulase positiva e B. cereus.

ABSTRACT

Cronobacter sakazakii é patogênico, importante contaminante de fórmulas infantis, que apresenta tolerância aos sanificantes comerciais. Assim, os óleos essenciais se destacam. O objetivo desse trabalho foi avaliar a adaptação homologa e heteróloga de células sésseis de C. sakazakii aos óleos de canela, ho wood e menta. C. sakazakii foi adaptado a concentrações subletais dos óleos e desafiado a diferentes concentrações dos mesmos, determinando-se a adaptação homóloga. Já para avaliação da adaptação heteróloga os óleos foram diferentes daqueles usados na adaptação. De modo geral a adaptação tornou as células sésseis mais sensíveis aos óleos, entretanto, houve adaptação homóloga para o óleo de canela. Também foi observada adaptação heteróloga. C. sakazakii na forma séssil foi capaz de desenvolver adaptação homóloga e heteróloga.

ABSTRACT

Staphylococcus aureus é uma bactéria patogênica, associada à toxinose alimentar e pela sua capacidade em se tornar resistente a grande número de antimicrobianos. Alternativa para seu controle é a utilização de cinalmadeído, componente majoritário do óleo essencial de canela. O objetivo desse trabalho foi avaliar a inativação S. aureus por cinamaldeído em duas temperaturas. S. aureus foi incubado em caldo triptona de soja adicionado de 0,5% de Tween 80 (v/v) e 0,25% (v/v) de cinamaldeído nas temperaturas de 37 °C e 7 °C por 5, 10, 20, 30 e 35 minutos. Não foi observada ação bactericida do cinamaldeído na temperatura de 7°C, entretanto, a 37°C, após 35 minutos de contato não foram detectadas células viáveis. Dessa forma, destaca-se a importância da temperatura para a ação antimicrobiana do cinamaldeído.

The essential oil (EO) extracted from the bark of Cinnamomum zeylanicum (Czey; also known as cinnamon), mostly derives its properties from its major compound trans-cinnamaldehyde (TCin). The present study evaluated the antimycobacterial activity of the essential oil from Czey (CzeyEO) and TCin against sensitive and resistant clinical isolates of Mycobacterium tuberculosis, as well as the combinatorial effects of CzeyEO and TCin with the anti-tuberculosis (TB) drugs rifampicin (RIF) and isoniazid (INH). The resazurin microtiter assay method was used to determine the minimum inhibitory concentration (MIC) of the components tested on the clinical isolates of M. tuberculosis. The effects of the CzeyEO/RIF, CzeyEO/INH, TCin/RIF, and TCin/INH combinations on the M. tuberculosis H37Rv reference strain were evaluated using the checkerboard method to determine the fractional inhibitory concentration index (FICI). CzeyEO and TCin inhibited all bacterial clinical isolates. In the interactive experiment, CzeyEO and TCin were found to be highly effective in reducing the resistance of resistant M. tuberculosis to RIF and INH. All four tested combinations demonstrated synergistic and additive effects, with no antagonistic effects. The synergistic combinations of CzeyEO/RIF and CzeyEO/INH exhibited FICI values of 0.375 and 0.5, respectively, while the TCin/RIF and TCin/INH combinations exhibited FICI values of 0.31 and 0.5, respectively. These results indicate that CzeyEO and TCin are potential candidates for the treatment of drug-resistant tuberculosis in combination therapy with INH and RIF.


O óleo essencial (EO) extraído da casca do Cinnamomum zeylanicum (CzeyEO), conhecido como canela, tem como seu principal composto o trans-cinamaldeído (TCin). O presente estudo avaliou a atividade antimicobacteriana de CzeyEO e do TCin contra isolados clínicos sensíveis e resistentes de Mycobacterium tuberculosis, bem como os efeitos das associações de CzeyEO e do TCin com os fármacos anti-TB, rifampicina (RIF) e isoniazida (INH). A técnica de ensaio de microtitulação da resazurina foi utilizada para determinar a concentração inibitória mínima (CIM) dos componentes testados nos isolados clínicos de M. tuberculosis. Os efeitos das associações CzeyEO/RIF, CzeyEO/INH, TCin/RIF e TCin/INH contra a cepa de referência H37Rv de M. tuberculosis foram avaliados pelo método Checkerboard, determinando o índice de concentração inibitória fracionária (ICIF). Todos os isolados clínicos bacterianos foram inibidos por CzeyEO e TCin. As interações de CzeyEO e TCin foram altamente eficazes na redução da resistência do M. tuberculosisresistente a RIF e INH. Todas as quatro combinações testadas resultaram em efeitos sinérgicos e aditivos, sem efeito antagônico. Ambas as associações de sinergismo de CzeyEO/RIF e CzeyEO/INH mostraram valores de ICIF de 0,375 e 0,5, enquanto as associações de TCin/RIF e TCin/INH apresentaram valores de ICIF de 0,31 e 0,5. CzeyEO e TCin são potenciais candidatos em terapia combinada com INH e RIF para o tratamento da tuberculose resistente.

With the objective to evaluate the efficiency of essential oils of Citrus latifolia (Tahiti lemon) and Cinnamomum zeylanicum (cinnamon bark) in the control of plant pathogens Penicillium sp. and Aspergillus sp. and the quality of the bean seeds, two experiments were conducted. In the first one, the effect of essential oils of C. latifolia and C. zeylanicum was evaluated in vitro development of the fungi Penicillium sp. and Aspergillus sp. and, in the second one, the influence of essential oils on the physiological and sanitary quality of bean seeds. The variables mycelial growth, conidial germination and sporulation of Penicillium sp. and Aspergillus sp. were measured in the first experiment, while the seed germination test, first count of germination, germination speed index (GSI) and sanity test of bean seeds were measured in the second. The essential oil (EO) of C. zeylanicum was more efficient than C. latifolia in the control of Aspergillus sp. and Penicillium sp., but decreased the physiological quality of the beans seeds. The fungal diversity identified in the seed health test was composed by fungi of the genera Aspergillus, Penicillium, Cladosporium, Fusarium, Chaetomium and Rhizopus. The results indicate the potential of the use of these EOs in the seeds treatment.(AU)


Com o objetivo de avaliar a eficiência dos óleos essenciais de Citrus latifolia (limão taiti) e Cinnamomum zeylanicum (canela em casca) no controle dos fitopatógenos Penicillium sp. e Aspergillus sp. e na qualidade das sementes de feijão, foram conduzidos dois experimentos. No primeiro, avaliou-se o efeito dos óleos essenciais de C. latifolia e C. zeylanicum no desenvolvimento in vitro dos fungos Penicillium sp. e Aspergillus sp. e, no segundo, a influência dos óleos essenciais sobre a qualidade fisiológica e sanitária das sementes de feijão. As variáveis crescimento micelial, germinação de conídios e esporulação de Penicillium sp. e Aspergillus sp. foram aferidas no primeiro experimento, enquanto o teste de germinação de sementes, primeira contagem de germinação, índice de velocidade de germinação (IVG) e teste de sanidade de sementes de feijão foram aferidas no segundo. O óleo essencial (OE) de C. zeylanicum foi mais eficiente que C. latifolia no controle dos fungos Aspergillus sp. e Penicillium sp., mas diminuiu a qualidade fisiológica das sementes de feijão. A diversidade fúngica identificada no teste de sanidade de sementes foi composta por fungos dos gêneros Aspergillus, Penicillium, Cladosporium, Fusarium, Chaetomium e Rhizopus. Os resultados indicam o potencial do uso desses óleos essenciais no tratamento de sementes.(AU)
